The flights will operate on a weekly basis and allow winter sun seekers in Northern Ireland to have a greater choice of destinations.
Jet2.com's boss Philip Meeson said the new route reaffirms the airline's commitment to "offering customers even more choice when it comes to their holiday destination".
"We strive to provide only the best and Tenerife gives some of the best year round sun to help people get through the winter months," he said.
Mr Meeson added that he expected a "huge influx" of passengers on the flight to the Canary Island, which is expected to prove popular among both families and couples.
Tenerife boasts the popular beach resorts of Playa De Las Americas and Los Cristianos and is famed for its volcanic landscape and whitewashed villages.
Leeds-based Jet2.com operates flights to more than 40 European destinations from its six UK hubs.
